Fastest route to 1 Billion ISK: 2 characters ratting for 12-16 hours straight in good true sec drone space. The 12-16 hours includes the time to prune the system. Disadvantage here is that if hostiles show up, all that time spent pruning can be wasted. Despite that, when I lived in P3X (dead end system, -.91 true sec, bubbles on the gate, 19 belts), I could easily make a billion a day (12-16 hours) ratting with 2 marauders. That was after paying a 15% refinery tax to alliance and selling minerals to corp at 5% under Jita price.
For missions, it depends a lot on social skills and how savvy you are about spending your LP. If you get good LP:ISK conversion, it doesn't take that long. If you figure 7500 LP per mission, which is perfectly manageable with a high sec Level 4 agent, and a minimum of 1000 ISK:LP (you can usually do better than this with market research), then it takes about 133 missions to make 1 billion ISK worth of LP. That many Level 4 missions will of course also net you a ton of rewards, bounties, loot, etc. So, figure it only takes 75 missions to get 1 billion ISK (being very conservative here, might be less than that). Some missions are worth looting, others not - knowledge comes with experience.
How many missions can you solo in an hour? It will depend on the missions of course, but with my two good mission runner accounts (one Navy Mega/Kronos/Domi + one CNR/Golem/NH), I'd count on being able to do 75 missions in 25 hours (not consecutively - I'd be suicidal). For solo mission running, with a bog-standard Raven, I'd say more like 50 hours minimum. The advantage of two accounts is that you have more missions to choose from so you can skip the bad ones, in addition to the immense amount of time you save by faster killing and never having to warp out at all.
Timing is also important. Prior to Apocrypha, I piled up close to a million SOE LP on my various characters. I turned that into over 3 billion ISK (conservatively) within a couple of days. That market is no longer nearly as good as it was then, but with some careful planning, absurd returns on mission running time are possible.
Note that all of these ISK-making ventures require serious investments of time, energy, SP and capital.
